Here's the instructions that came with mine:

1 Submerge the entire hose and gravel tube into the aquarium water. Manipulate the unit so that all interal air escapes and the entire unit fills with water.

2 Hold your thumb tightly over the end of the hose so water does not escape. Lift the end of the hose that you are holding out of the aquarium and lower it into the bucket(the bucket should be below the bottom of the aquarium).

3 Remove your thumb from the end, and water will begin to siphon from the aquarium.

4 Immediately hold the cylinder vertical and begin vacuuming the gravel using an up-and-down motion. Some gravel will rise into the gravel tube, but it will fall into place as gravel tube is lifted.

I would not recommend using it to remove water from your tank. You need to turn on the tap and it wastes a horrible amount of water and does not save you much time. Continue to use the syphon-and-bucket tube to remove the water from the tank.
To add the water with the python, attach the screw end to your kitchen faucet (or whichever tap). You may need to unscrew the current fixture at the end of the tap, and you may need to try several different areas to get the write fit for your tap. When you do, screw it on. You should be able to open the bottom of the screw part to let the water run out. Let the water run out until it is the same temperature as the water in your tank.
Then put the other end of the hose in your tank. You may need to close the lid on the hose because the pressure in the hose can make the hose fall out of the tank. Also put the amount of water conditioner that you need into the tank water.
Then close the screw part so the water runs into the hose and into the tank. Let it run until the tank is full, periodically checking the temperature to make sure it's still the same as the tank's temp.

Its a venturi pump. If you google venturi waterbed, you'll come up with a bunch of information on how.

Basically its a funnel with a hole cut in the side. water passes through the funnel & it creates suction in the side hole. Its not much suction either. Just hook it up to your sink & attach the hose to the side thing. Put it in the tank & turn the sink on... it should start draining the tank.

Its pretty inefficient compared to just using a gravel syphon or a mechanical pump.
