Do i have to use the quadrant method or can i spread the substance on all of the the petri dish

im talking about inoculating the medium, but will i still see colonies even if i dont use the quadrant method

You may or may not see colonies if you don't use the quadrant method. Depending on how concentrated the bacteria are, if you just spread it out over the entire plate you will possibly have a "lawn" of bacteria, or just a solid mass over the whole plate, in which you will not see individual colonies. Realistically, you will probably still be able to see a few individual colonies at the sides of the plate.

The idea behind the quadrant method is that as you sterilize the loop each time you are dragging fewer bacteria across the plate and essentially diluting them so that in the final quadrant you have just a few colonies all nice and isolated so you can pick them easily.

If you are talking about the medium, generally it is spread all over the dish. If you are talking about innoculating the medium, the quadrant method allows one to see individual colonies better and select from the colonies for further tests.

I prefer to just put sodium in them and add water.
