No. Usually this expression is used of a team in a sports playoffs (for the championship). If refers to a team getting in only to lose the very first game and so be done --out of the playoffs. (It's NOT something any team likes!)

Hmmm, I've also heard this used by people who have one child and are asked if they are planning to have another. "One and done," here means "no," we will not be having any more children that we can foresee. There may be an implication that the process of bearing, birthing and raising one child is difficult enough--but there is no suggestion that it wasn't "worth it" :) It's always worth it! My two cents.
