Well if the other guy is dating your sister, he's obviously not gay, so to protect your gay friend, I wouldn't tell him. Chances are the guy will laugh at him and make jokes about it forever.

If he's dating your sister, he obviously likes girls and wouldn't be interested. I don't see what the point would be in telling him. It might just embarrass him and ruin their friendship (if they have one).
And, on the off chance you sister's bf is bi and decides to leave your sister for him -- wouldn't that kinda hurt her feelings? ;)
