Well, if you're asking in terms of the best film, it is by far Grave of the Fireflies. This is the supreme example of symbolism in a film. It explores human nature. One, that adults have far much more to worry about than to protect the youth of the nation. Two, that a young boy's pride can unknowingly be the worst choice of his life. Three, that a young girl can put her life into her brother's arms, and that trust, however destructive, is the most comforing place you can go. There are many more reasons, but I'd suggest watching the film. The only downside is that it's quite depressing for those who prefer to watch light-hearted anime such as DBZ or Pokemon ( both of which could never measure up to being considered "the greatest anime of all time". I'm sorry but DBZ and Pokemon have no substance whatsoever.)
If you want a series.... then I suppose Vision of Escaflowne. It's a breathtaking roller-coaster the entire series and smashes into a refreshing halt as it ends. A bit too short in my opinon, but the fact is that it gets to the point and avoids lollygagging around with trivial matters overrules the length. Sometimes trivial matters can be amusing to dwell on, but it's obvious just by this anime that miniscule plot twists are pointess and unnecessary. The low of this anime is that those who like watching anime that span over 100 episodes in length will not like this. Their calling would be that of Maison Ikkoku, which is also excellent.
Hopefully some (read: definitely everyone!) will agree with me because said film and series possess a level of merit that surpasses all others, in my personal opinon. I have given my reasons and believe I am right, but I do suppose that everyone has their own opinion.

Neon Genesis Evangelion!!!!!!!!!!!!! Practically brought anime back to life... It's awesome but I guess it was kinda short and the endings are kinda confusing, but still It's probably the best anime ever!!!!!!!!!!!!

I know many people love LONG running Animes like DB and Gundum, but I think my favorites are Case Closed, and Fullmetal Alchemist. I think the movies Akira, Howling Castle?(I think that is the title) and Ghost in the Shell could be considered the Greatest as they made allot of people take notice of Anime or Japanimation as I knew it.

Yu Yu Hakusho and Ruroni Kenshin. I love the story of Yusuke Urameshi( Yu Yu Hakusho) and Batousai the Man Slayer( Himura Kenshin)! I loved watching these animes on Toonami!

i would have to choose Trinity Blood.
itz the greatest cause there's so much blood in it!
itz also good cause it has a good story line.
the artwork is prety amazing too!
if you watch it than you should like it too
