Everytime I have conversations with him I can see that he is very uncomfortable, around me. How can I get him to realize that I am not interested in him.

You may be misreading his discomfort. He may just be awkward socially.
You have a few choices.
1) Avoid him, if you are uncomfortable around him (this isn't very helpful to either of you, imho)
2) Ignore the situation (also not very helpful)
3) Deal with it directly. Say something like, 'I notice you seem uncomfortable around me. I want you to know that I think you're a great guy, but I'm not attracted to you. I'm not gay. I hope I'm not assuming too much by saying this.'

If you just don't want to tell him which is the best way tell him what kind of girl you are interested in. Maybe he will get the hint.

Be honest and say "thanks but no thanks", no need to be aggressive or rude. Just simply tell him that is not your preference and no harm done.
