if she is your best friend you should be able to just take her to a private room and talk to her about your feelings and she should be able to understand why your asking her this.. I know how you feel because that happened to me but once I talked to her about it everything was OK so just talk to her and good luck

put on some sexy lingerie and prance around her. offer to give her a lap dance. rub up on her if she says yes while you are giving her the lap dance. you should be able to tell if she is gay or not if you do that.
or you could just leave it alone

if you're best friends you should be comfortable talking with each other.
just talk to her about it.
in private.
think about why you think she is. and maybe ask her how she feels about people?
but don't jump to conclusions. she could just be curious
