The largest by VOLUME is Boeing's Everett Plant near Seattle, Washington, 13.3 million cu. meters (470 million cu. feet). But of course, most of that is just "air space".
The largest by FLOOR AREA is The Merchandise Mart in Chicago, 390,000 sq. meters (4.2 million sq. feet) of space you can walk around.
The TALLEST is The Sears Tower, also in Chicago.

Currently, the Sears tower in Chicago holds the honor. However, it is due to be knock from it's top position by the Venetian/Palazzo hotel mega-complex in Las Vegas when construction finishes on the Palazzo sometime this year.
