You need to wait for your iTunes program to say that it has completed the update. Then eject the iPod from iTunes (there's a little icon to the right of the iPod's name in the left menu bar). If you really feel like being safe, eject it from your computer next (icon with a green arrow in the system tray). But depending on your setup, waiting for your iPod to tell you it's safe to disconnect could leave you waiting for a very long time. When I first set mine up, it would always display the Do Not Disconnect screen until I ejected it from iTunes (then it would switch to the charging screen). Now, as soon as it's done updating, it'll switch back to the main menu screen until it times out to the charging screen, but I no longer need to eject it from iTunes (it still shows up in the list on iTunes, but it vanishes from My Computer as a readable drive).
