It is never really that safe to work on the roof of a house, Falls from heights are the most common type of accident on construction sites and therefore there are many precautions taken in the construction industry.

Most of the techniques used will probably not be possible to carry out yourself, things such as using a guard rail around the perimeter of your roof.

I would suggest using a harness (similar to one you might use for rock climbing) and attach yourself to something very firm on the roof of the house. You might be able to rent a harness from a tool hire shop and they would be able to give you some training and guidence with.

I would either call a tree service to remove it with their crane/boom OR if I felt really lucky, brace it at least -3-4 places between the tree trunk & the garage, then start at the tree and cut it off with a chainsaw in manageable size sections. Once you get it cut down enough, you can remove the branches that penetrated the roof and remove it completely.

you did not state a flat roof or a pitched roof for a pitched roof scaffolding and cat ladder for the level roof it should be alright to stand on

It is never really safe to work at heights. But you could get scaffold or use a roofers ladder.
If you are really worried, I would suggest that you get in a professional

From ground, call a local contractor appropriate for the work that needs to be done.

all depends on what roof it is and what your doing?
slate roof cat ladders
all tiled roofs kick tile up and use battons as ladder

don't lean over the edge.
roof jacks
harness with attaching point nailed to peak rafter.
scaffolding on side or edge of roof
manlift

tie yourself to the chimney with bungy ropes then if you fall down will just bounce back up
