You've already got a lot of good recommendations for anti-spyware programs in the answers so far, so I'm not going to add any more.

Just one thing, though: If it is a pop-up browser window alerting you that you have a spyware problem, and the pop-up is advertising an antispyware or security program, DO NOT buy what the pop-up is selling!! Any program advertising like that is most likely either ineffective, misleading (for example, scans for free then tells you it costs to clean), or is itself spyware.

The best page I found about spyware (and viruses & adware) is the one listed below from ez computer tips. It has worked every time I have used it, even for the bad ones. If you already have anti-virus software then don't download the one they recommend. Two anti-virus programs on your computer is a BAD thing. Just follow the instructions and it will save you a trip to the computer repair shop.

The best defense against spyware is downloading and using Firefox instead of Internet Explorer.

FYI: Internet Explorer is the biggest reason people get spyware.
