1.The wireloop is usually dipped in ethanol. Thus if you are not careful there is the risk of fire

2.Depends on the media. E.g. LB is autoclaved. If you need to add vitamins, antibiotics or other heat labile substances then you add those after autoclaving and cooling the media. The solutions of those compounds that will be added to the media need to be sterlized by filtering with a 0.22 micron filter.

3.I guess you mean more effective sterilization process. Autoclaving of course.

autoclaving / retorting is more effective than boiling.
Boiled food still contains bacteria, autoclaving doesn't.
