No. De-Spell is a Spell Speed 1. You can only respond to your opponent's actions with Spell Speed 2 or higher. Quick-Play Spell Cards are Spell Speed 2 and are recognized by the lightning bolt symbol on them. Mystical Space Typhoon and Book of Moon are examples of Quick-Plays. Counter Traps are Spell Speed 3 and have a curved arrow on them. Counter Traps can only be chained to by other Counter Traps. Seven Tools of The Bandit and Solemn Judgment are examples of Counter Traps. Note: As a general rule of thumb, if a card doesn't say it negates AND destroys Spell or Trap card you can't use them in that capacity.

De-spell can be used on continuos or equip spell cards. If you take the time to read the rules or some advanced gameplay, you'll discover the difference between negate and destroy. And for your question, No. De-spell can only be used during your turn. If you want to negate and destroy powerful spell cards like Monster Reborn? Try adding Spell Shield Type-8, Magic Drain, or Magic Jammer to your deck.

Unfortunately, De-Spell only works on your turn, which means that you couldn't activate it anyway. This is the card description:
De-Spell
Destroys 1 Magic Card on the field. If this cards' target is face-down, flip it face-up. If the card is a Magic Card, it is destroyed. If not, it is returned to its face-down position. The flipped card is not activated.
I'm sure that you know that and all, but I'm just putting it here in response to the first answerer. Because De-Spell is not a quick play Magic Card, it can't be played from your hand. As stated before, it can only be played on your turn.
A better card to use would be Magic Drain (negates the activation of any magic card unless she has a magic card in her hand to discard) or Magic Jammer (where you discard one card from your hand to negate the activation of a Magic Card).

Yes... you can quick-play de-spell, so you can do it quickly enough to take out monster reborn before it activates. Also, you can play it right from your hand even if it's not your turn to activate it to cancel out monster reborn. Remember that next time you don't want to face the Dragon xD

de-spell is not a quick play magic card so you have to wait until it is your turn to activate it but you could use magic jammer
