Slappy McStretch answer is about as close an answer as you can get. They're are too many to choose from, Hendrix was awesome, and there are tons of great guitarist in all generations. Tom Morello from Audioslave is the new Eddie Van Halen. But since I am a Texas Boy and you probably want one answer i'm going to say Stevie Ray Vaughn. Because he shred. But other than that their are just too many.

There's so many different sub-genres of rock.
If it were pure guitarist - Jimi Hendrix
Blues-rock and classic rock - Eric Clapton
Hard rock - Jimmy Page.
Heavier rock, a precurser to metal - Tony Iommi of Black Sabbath
Progressive rock - David Gilmour of Pink Floyd
