That is known as the click of death. It is a sign of a dead or dieing hard drive. The only other possibility is that the click is coming from your Power Supply. Occasionaly there will be a click if a surge hits the PSU. Have a professional test both to see which is the culprit. Any professional can tell instantly where the click is coming from.

My bro had something like this happen except when it was turning on. He said it was an operating system failure. He had to use linux to start up his computer and get his data, burned it to a dvd, & then reformated his entire hard drive. If you're not computer savy, I'd suggest taking it to a computer store. I have to ask my bro & dad for help!! lol I'm not sure this is the same thing as you have, but if it isn't, it's probably the hard drive. Hope this helps!!
