We did a similar trip on our honeymoon.
It takes about 6 hours to travel from Orlando to Miami itself.
Stop in Fort Lauderdale on route if you go down the east coast. It's not too far from Miami but it's called the Venice of America as it's all canals etc.
When we did the trip we did the west coast. I would definately stop in Fort Myers and Sanibel Island. Sanibel Island is gorgeous. it's vey similar to Key West, all quaint and caribbean painted colours. Amazing!
As for fitting in the Keys it is do-able from Miami depending on where you go.
Miami is about a 4 hour drive from Key West the furthest key. Now if you are willing do do that journey make sure it's not on a weekend as the traffic will be horrendous as there is one road in and one road out of the whole of the keys.
If you go to Key Largo or Duck Key it's a bit more manageable to drive - couple of hours tops from Miami.
If you have to travel back to Orlando to get home I would suggest getting a short flight from key West or Miami. A single flight from Key West will set you back about £100 (flights with Delta and Continental airlines) / Miami to Orlando about £50 (United, Delta, American and Continental flights).
You can arrange for your car hire to start at Orlando and drop back at either Key West or Miami airport. We did that. Check with different car hire companies as most won;t even charge you a one way drop off fee (we used Avis).
I personally didn't rate Miami as much as Key West.

The keys are really far from Orlando. We were going to do it one year and then decided just to do the gulf coast instead, because we would have spent most of our time in the car.
On the gulf coast, I would see Fort DeSoto Park. It's really beautiful, near Tampa.
St. Armand's Key near Sarasota is a cute place for shopping, Sanibel Island near Ft. Myers is very nice as well.
Then, you can proabably do one of those boat tours of the everglades on your way across the state to Miami.
If you are going to do the keys, you probably won't have time to do Key West, you will probably only be able to do Key Largo or Marathon.

Tarpon Springs. Just north of Tampa. A quaint town with some incredible Greek restaurants. My fiance and I live in Orlando and we travel there once at least once a month. Make sure you check out the fish specials at Hellas Restaurant.

I think you'd be pushing it to do it in 6 days. Placed to stop: Sanibel Island, Naples, various points through the Everglades.

do not miss out the keys, and make some time for them. Some of the best views and memory's for me . As for the rest i would not know where to begin. just so much to take in.

definately stop in St.Petersburg-there is a gorgeous harbour area and the Dali Museum which is amazing!
Everglades are a must too!!
