Most of the EM techniques like Cryo EM study microtubules in solution not in cell samples. I wanna see it in the cells. What about freeze fracture deep etching EM? I would like to know about the solutions (name) what they do and the time of treatment. I mean a general steps of the experiment if possible mention the equipment used for this.

Is it possible to still see microtubule catastrophe if stained with immunogold?

Why don't you use tubulin antibody, and look at it with fluorescent microscopy?

(you'll have to excuse me, but I'm not really sure what you mean by microtubule catastrophe...)

You'll need to treat the cells somehow. I'm not an EM guy, but i work alot with colcemid, which does the job nicely.
