What makes a book a science fiction book?

The simplest answer is that Science Fiction requires an extrapolation of science... such as space travel or alien lifeforms... and that is at least theoretically possible in some future science. If the story requires magic, then it is Fantasy. There are a couple exceptions. It is virtually universally believed that it is impossible to travel faster than light or to travel back in time, but those have been accepted as Science Fiction for a long time now. On the other hand, there is some evidence that werewolves are possible but they have been part of fantasy forever.

Because it involves an advancement of science, virtually all science fiction takes place in the future but that is not a requirement. Alternate Reality stories set in the past are also considered science fiction. Harry Harrison's "West of Eden" stories which involve early man dealing with technologically advanced dinosaurs are a good example as is Norman Spinrad's "The Iron Dream" in which Adolf Hitler is a science fiction author.

It is a broad genre. A typical Science Fiction book will have spaceships, clones, robots etc. and is usally set in the future. This genre sometimes will cross over into Fantasy.
