Why would you want to anyway? If you're using the upgrade disk then before you install it, type at the command prompt:
convert c: /fs:ntfs
Reboot and let the conversion run. When done, install Vista. Unfortunately Vista upgrades are not like it's predecessors where you can do a fresh clean install with the upgrade disk by briefly inserting a previous version when prompted. Vista upgrades have to have XP already installed on the machine. So, to have the cleanest possible installation with the upgrade disk, then backup all your data then kill the partition and install XP and let it create the ntfs partition for you. Then chose "quick format" and let it continue. When done, don't install any drivers or updates, but just go straight to the Vista upgrade disk. When done, then install your drivers (if needed) and all your updates.

The quick answer is if you are upgrading Vista will convert you Hard drive to NTFS (no loss of data). However, if you have a program that specifically needs FAT32 then you are out of luck.

Why do you need know if it can?

NO. Vista works on NTFS but its backwards compatible.. why are you running FAT32 anyways? its less secure. You need a robust technology like NTFS. NTFS (New Technology File System).
