Koch's postulates is used as a method of linking a disease to a microrganism and where developed by Robert Koch and Friedrich Loeffler in 1884 then improved by Koch again. To prove the link
1.The organism must be found in all animals suffering from the disease, but not in healthy animals.
2.The organism must be isolated from a diseased animal and grown in pure culture.
3.The cultured organism should cause disease when introduced into a healthy animal.
4.The organism must be reisolated from the experimentally infected animal.

The second part of the first postulate was later dropped because so people can carry illnesses without being symptomatic (such as tb).
hope this helps
