Ah, this isn't as easy a question as you'd think, there's at least 3 possible answers.
1. In the original Star Trek series, the first captain we encounter is James T Kirk, played by William Shanter
2. In a later episode of Star Trek (the Cage), we are introduced to a previous captain of the enterprise. Christopher Pike, played by Jeffrey Hunter
3. Finally, in the latest spin-off, Enterprise, which tells the tale of the very first starship Enterprise, the captain is Captain Jonathan Archer, played by Scott Bakula
So, it depends on if you are meaning chronologically in the Star Trek universe, or ours I guess. There's also an animated series, where there's a captain that predated Pike, but not Archer (I think he's Captain April) but a lot of Trekkies don't take the animated stuff too seriously.

Scott Bakula since his Enterprise is the first starship bearing that name in Starfleet.
However if you mean first on screen then William Shatner (Jeffrey Hunter as Christopher Pike was first only seen in the flashback episode The Menagerie as the original pilot The Cage wasn`t shown until the show was established)

The first Star Trek captain is Christopher Pike, who commands the USS Enterprise during the Star Trek pilot, "The Cage". However, in the Animated Series it states that Captain Robert April preceded Pike as commander of the Enterprise, but this has never been made official by the producers

Appeared in the original Star Trek pilot "The Cage" and the episode "The Menagerie, Part I".
Best known as commanding officer of the U.S.S. Enterprise NCC-1701, just prior to James Kirk, Pike counted the top-secret first contact with the natives of Talos IV amid his long Starfleet career. Later in life, as a fleet captain aboard the training ship U.S.S. Republic, Pike suffered massive radiation damage after saving several cadets when a baffle plate ruptured in the ship's reactor. Thanks to the unorthodox methods of his former science officer, Spock, in 2366, Pike was returned to Talos IV to live out his life without the debilitating lifestyle he had endured, even with a self-propelled, life-support wheelchair.
Note: Captain Robert April (The character has not appeared or been mentioned in any live-action Trek works, but has made frequent appearances in spin-offs. For example, he is listed in the Chronology as the first captain of the Enterprise, accompanied by a photo of the creator of Star Trek Gene Roddenberry, sitting on a captain's chair wearing a Starfleet uniform, and is listed as such on the official Star Trek website) preceded Pike as commander of the Enterprise, if you follow Star Trek lore, Thusly would had been the first Captain of Enterprise NCC-1701. Current lore cites Captain Johnathan Archer, played by Scott Bakula as the First Captain of the first Starship Enterprise, Which is pre-federation and pre NCC-1701.

The Star Trek: The Animated Series revealed a previous captain, Robert April. Although the animated series is not recognised as official canon by Paramount, many other spin-off works recognise April's first year mission. (It has been reported that April is identified as the first captain of the NCC-1701 on a computer display visible in the Star Trek: Enterprise episode "In a Mirror, Darkly", however this has not been verified.)

Jeffrey Hunter played he first officially known commanding officer of the Enterprise, Captain Christopher Pike, who served as captain for more than a decade prior to James T. Kirk.
In episodes with footage of an older Capt. Pike, the character was played by Sean Kenney.
