The reason that human (acutally all mammalian)mature red blood cells lack a nucleus appears to be so that the red blood cell has room for more hemoglobin and therefore can carry more oxygen per cell.
Remember the human red blood cells doen't always lack a nucleus. Up until the final steps of red blood cell maturation, they do have one.

The short answer is that certain cells are not required to reproduce and have no need for a nucleus containing DNA. The DNA is contained in the stem cells that reproduce the erythrocytes. Brain cells also do not usually reproduce but may under certain limited circumstances to repair damage. Heart muscle also does not usually reproduce except to repair damage (like a cut finger).
