The Cretaceous-Paleogene transition (about 65 million years ago), when an asteroid strike in the Yucatán peninsula (now Mexico) caused the extinction of the dinosaurs and allowed mammals, and therefore much later humans, to become the dominant land species.

To be quite honest, the birth, life, and death of Jesus Christ. Think about it, originally, we changed from B.C. (used to stand for "before Christ") to A.D. That alone distinguished the way we date time and history, which is pretty influential. And, whether he was a man or God or both, the entire world acknowledges that he once existed, yet the entire world cannot agree to what he really did while on this planet. He changed how people view heaven and hell, and how to get to one of them based on your life. Just say his name, and you'll stir up all kinds of controversies wherever in the world you go. That's pretty globally influential if you ask me (which you did!).
