Your brother and others in the family are just reacting to a situation that they can't understand. I wonder where you live - I live in a big, cosmopolitan and student-orientated city and I can tell you, it is very common for girls and gay guys to be best friends - in fact I would suggest it is quite a fashionable thing to do!
Rest assured that there is absolutely not a thing wrong with this situation and continue to enjoy your friendship. Ignore the family comments - that's what families are all about!

Most of the responses you're getting from your housemates are probably because it's a unique situation for some of them, and they don't fully know what to think about it, so they resort to trying to make it into a joke. On the other hand, it wouldn't hurt to take a step back and re-examine yourself for a moment, even if the result is that you find out that the people in your house are way, way off the mark.
If you're not dating anyone right now, that's probably adding fuel to the fire by causing them to see emotions that might not actually be there. If after some serious introspection, you discover that you do feel some sexual attraction to this guy, your housemates might be picking up on this. It's pretty usual for feelings like this to develop, and you'll want to be sure to keep them in check, since it could end up hurting you emotionally over the long term.
If, however, you find that you have absolutely no sexual attraction for your friend, then you can rest assured that your housemates have no clue what they're talking about, and you can just write off their opinion. At a minimum, though, you should probably talk to your friend about this. If nothing else, it will give you two a good inside joke to chuckle about, which will help make the whole situation easier for you.

Yeah my advise to you, is to tell your family to smarten up and quite being so childish and homophobic!
In our society when somebody refers to their 'boyfriend' it is implied that the two people are in some kind of sexual relationship. It's OK for a girl to have a 'girlfriend' and this does not imply anything sexual but if somebody has a 'boyfriends' be them a str8 girl or a gay guy then something sexual is implied. If these guy you call your boyfriend is truly gay he would not be sexually involved with you because if he was and if you know for a fact that he also liked to be sexually involved with another guy then he would be bisexual not gay! And if you two are not sexually involved and you have not intention of ever becoming sexually involved then I would say (under my terms of 'boyfriend') that you do not have a boyfriend in this guy! You can remain friends with this guy but if I was you I would be getting out and meeting new prospects for a boyfriend ta ht can love you like a women!

if that is all these people have to make your life unpleasant than consider yourself lucky.
do you really care what they all think?
start firing stuff back at them, give back what they say to you when their friends call. make them feel uneasy and unpleasant when their friends are around.
besides, it is a brother duty to give his sister a hard time, and it is your rightful duty to do the same back to him. it's all part of growing up in a family with brothers and sisters.
