It depends on the game. If it's a game like Worlds of Warcraft, then yes, you need to be online to play the game. At the bottom of the box (or in the area marked system requirements if you're buying online), you'll see all the items needed to play the game. If it includes an internet connection, then you most likely need to have one to play the game.

Is it a mmorpg game? If it is then you need internet connection. Such games is Counter Strike, Warcraft etc..
