The best solution is to backup everything and reinstall. (it's a good idea to backup frequently anyway) The "clean sweep" style programs don't usually do a good job.
Often, just upping the RAM can breathe new life. If you have less than 512MB and running XP or Vista it's a near necessity - and usually really cheap.

First of all, I suggest you to uninstall the programs that you don't need. After that, you must use CCleaner (CrapCleaner) http://www.ccleaner.com/download/
I think it must be used by every Windows user and it is very easy to use.
It will clean all of the Internet browser cache, temporary folders and files and similar files that waste your disk space. Also it has the capability to clean the windows registry from the wrong and useless entries of uninstalled softwares.
Lastly, run Defrag for your hard disks.
I suggest, do these jobs and also antivirus scan at least every month.

Then restart your computer.
This is the simplest solution that should work.
