Without wanting to sound like a "geek" .. I would say "The Lord of the Rings" by J.R.R. Tolkein... it's beautifully written and has romance, intrigue, adventure, a huge cast of characters and the whole epic "good versus evil" thing going on.
Tolkein understood language, mythology and people. It is utterly engrossing and contains plenty of moral and ethical dilemmas.

There are so many great stories that it is hard to single out one and say that it is so superior to all the rest that it is "The Greatest". Personally, it would be easier for me to answer this question if it was a little more focused.
