I'm not sure what you mean by your question. Commonly, microscopes have 10X occular lenses, and so you'll have to have a 40X objective lens to give you a 400X view.

If you're talking about the specific steps, basically you'll lower the stage and put the slide on with a lower power objective in place. Then you'll raise the stage nearly all the way and then while looking through the microscope lower the stage until you can focus on the object with the lower magnification. Then, rotate the 40x objective into place and adjust the fine focus knob.

Huh. I could never get mine to work either. It seemed like the focal point was INSIDE the lense! I just assumed that my microscope was junk. Now I've got to dig it out and try that.
