I have a holiday booked to Grece with school and I'm really looking forward to it. But when i told work that I needed to book the week off my boss said she didn't think she could give me the time off because it'll be the easter rush so lots to do and she already has 3 people off. I lied, I told her I only found out that I was going a few days ago when she asked why i didn't tell her as soon as I found out, but I really just never got around to it, but I figured it'd be fine cos it's not till April. She's gonna talk to the manager about it, but says she can't promise anything. If they tell me I have to work, I'm gonna say they'll have to fire me though, I'm not giving up my holiday. I can't afford to, I'll lose what i paid for the holiday £650, maybe £150 if only the deposit, but that's still way more than a weeks worth pay. and I'm only contracted to do 6 hours a week, but i do get a lot of overtime. I'm not cancling my holiday to work, but if I don't can they really fire me?

Of course they can fire you. You can request time off, they can deny it, and they can fire you for not showing up. If you plan on going anyway, let them know in writing. And make sure you find another job to go to when you get back. Next time, let them know sooner. A week is a long time to need off.

They could fire you if you go when they tell you cannot have the time off. But I doubt it will come to that. If you are a good employee they will give you the time off, and at the same time they will probably give you a lecture about giving more advance notice when you want time off.

In the future, request your time off as soon as you know. Even if its months ahead. My boss has a calender and checks it for requests all the time.
Unfortunately, they can fire you. They may threaten it, but they may not actually do it if they really need bodies.

Give notice that you are quitting prior to your holiday. Quitting would look better for future employment than being fired. Probably they will become more accomodating if you give notice.
