Manchester is historically notable for being the world's first industrialised city and for the subsequent central role it played during the Industrial Revolution.
Similarly Osaka also rapidly grew industrially and earned the name Manchester of Japan.

I don't know much about manchester or the nickname of the manchester of japan, however from visiting Osaka a few months back, I was surprised by a few things, one, the people are definitely more outgoing and the food is very good and cheap down there.
If you go, make sure to hit Spa World if thats your thing, it was a relaxing day that I needed to get myself back in the swing for continued travel.
