It means you have one part of the substance in question and dilute it in 49 parts of the DNA Polymerase Buffer solution.
So if you use 1 mL of substance, you add 49 mL of solution to dilute it and weaken it/buffer it.
So you don't make solutions in China? It's similar to one part tea, 49 parts water= one damn good cup of tea.:)

"1 in 50" is a dilution factor/dilution ratio, which, in a general sense, you are being ask to make a 1 is to 50 dilution of your initial sample with a diluent (the solution you would use to dilute your sample).

so, if, for example, you want to dilute your DNA sample in a polymerase buffer with a 1/50 dilution, you would use, again as an example, 1 uL (microliter) of your DNA sample and dilute it with 49 uL of polymerase buffer...that should give you a 1 in 50 diluted DNA sample. The volumetric units could change depending on how much you are working on...I assume it's PCR, so that might be in micron volumes.

Hope this helps!
