There is no one best; they all do slightly different things. And you must be careful to get a genuine program from the original download site, because some people distribute fake programs that install spyware!

You need all these anti spyware programs;
http://www.ewido.net - AVG(Ewido) anti trojan/spyware.
http://www.safer-networking.org - Spybot Search and Destroy anti spyware. Use the Teatimer settings to protect Internet Explorer against browser hijacks, and the Immuniser to stop malware getting on your pc. Also you can use it to back up your registry.
Spywareblaster works with Internet Explorer and Firefox. It won't uninstall spyware that is already on your pc, but it will prevent hijacks and other malicious changes to your browser.

Plus;
http://www.lavasoft.com - Ad-Aware SE Personal anti adware. Adware isn't exactly spyware.

http://spywarewarrior.com/rogue_anti-spyware.htm - The Spyware Warrior site identifies fake anti spyware programs.

These are the 2 product I have come to rely on most. I volunteer as part of the SUPERAntiSpyware (SAS) malware hunters team because I support free products that are of such high caliber. Nick Skreptos, the maker of SAS is one of the hardest working guys in the business, constantly updating definitions and improving his product to detect/remove malware better. SAS will detect and remove Rootkits that some specialized Anti-Rootkit products won't touch. It's also quite impressive when it come to the Zlob/Smitfraud infections that display the fake security warnings. It's a must have on any system.

As for AVG AS, that is a longstanding and proven product since the days it was known as Ewido...enough said.

The fact still remains that even the best product will miss a certain infection on a given day, so it pays to have multiple "on-demand" scanners. Malware is released so frequently that it is impossible for any one product to catch everything out there
