No. Not at all, unless you might want to spend a day or two extra. The Trains from HYB to NGP travel via Secunderabad- Kazipet - Ballarshah- Nagpur. This route takes the Northern Andhra Pradesh and Eastern Maharashtra/Vidharbha route.
Kolhapur is in the South West of Maharashtra. You travel diagonally opposite to it.
For Mr Laughing Buddha:
Hyderabad- Kolhapur Haripriya link express exists. This has 1 AC 2tier, 4 sleeper class, 2 General and 1 SLR coach attached to the 7429 Rayalaseema express. These link coaches separate at Guntakal and travel via Hubli-Londa-belgaum to Kolhapur.

Never. But of course if the train is diverted for some reason you may perhaps. But if you want cover both kolhapur and nagpur, go by direct train from HYD to NGP and then catch Maharashtra Express from Nagpur to Kolhapur. There is no direct train or road route from HYD to Kolhapur. Please also see map of India

no way, both nagpur and hyderabad is in the central region of india and kolhapur is in the west coast .
