What are the advantage/disadvantage between Michaelis-Menten,Lineweav... Plot to get the Vmax and Km.?
i have read from internet, and know that Lineweaver-Burk Plot will suggest some inaccuracy in the determination of the Vmax and Km, but why we still use it, i think that it must have some advantage to use Lineweaver-Burk Plot. so need some one to tell me? sorry my english is not good enough.

The advantage of the Lineweaver-Burke plot is that you can draw it by hand using a ruler. Basically, people like straight lines when you can get them.

If you are using a computer to fit your data, there is no reason to use the Lineweaver-Burke plot. It is prone to inaccuracies due to improper error-weighting when you convert to reciprocal space. For example, let's assume your measurements all have approximately the same amount of error (which will often be true). In this case, when you do a curve fit, all of the points should be given equal weight. If you use a Lineweaver-Burke plot which uses the reciprocals of your measurements, this is no longer true. If you propagate your errors correctly, the error bars for your low concentration measurements will be huge. These points should not bear the same weight as the points with small error bars. A good scientific data analysis program (for example, Origin or Sigmaplot) can correct for this if you know how to enable that function; but you can avoid the problem entirely simply by using the Michaelis-Menten form.

I tried finding references on the web, but they are surprisingly rare given what a basic issue this is in the field of enzyme kinetics. I noted one in my source list. Technically, it's not referring to enzyme kinetics, but the math is the same. It points out that Scatchard plots have the same problem. Haynes-Wolfe plots are apparently okay though.

Either can be used, and you should be familiar with both. It depends on what information you know. Sorry, no good answer, it's almost a personal preference.
