Chargaff discovered that G content always equaled C content, likewise with A and T, meaning that purines and pyrimidines always paired together and so each pair was the same length. Then of course Rosalind Franklin's X-ray diffraction picture that showed an X shape indicating a double helix structure.

Don't forget Hersey and Chase showing that the material of heritability was DNA.

From the x-ray crystallography they easily deduced the mechanism of replication (but didn't actually state it in their seminal paper).
