The most usefull reason I can think of would be air changes per hour/day/week/etc. In other words, a 1000 CFM (cubic feet per minute) ventilation fan exhausting a 50' x 120' room with a 10 foot ceiling (60,000 cubic feet) has the capacity to provide one complete air change every hour. This is useful in many industrial applications where toxic fumes or combustible gasses are an issue (paint booths for example). Typically, these concerns are addressed at the engineering or architectural level and are considered by fire marshalls as well as other life safety professionals for occupancy ratings and fire suppression methods of buildings.

To work out:
1. The required capacity for a heater or air conditioner.
2. How many cockroach bombs to buy.
3. The size for a Halon fire extinguishing system
4. How much air you have in a sealed room.
5. Umm maybe how much lighting power you need?
6. Yes the ping-pong balls thing, were you ever asked it or wanted to play a prank on somebody.
7. How much stuff you can store in it (eg a warehouse).

This is a morbid reason but if a person was trapped in a sealed off room, knowing the volume of the room would allow you to calculate how much air they have to breathe and consequently how long they have to live.

Any time you intended to fill the space with something you need to know the volume, even if it is to filter the air in that room you'll need to know the volume of air you are to filter so you know the size(capacity) to purchase.
