The stomach, when full, is sealed by contracting muscles at the top and bottom of the bag and the stomach churns all the digestive juices and food together to digest it. It is then passed out of the stomach to complete digestion. Your stomach when it rumbles, i presume, is when there is no food in the stomach and the digestive juices are churning with nothing to break up and digest, this may cause the rumbling sound.

When you get hungry the acids build up in your stomach waiting for food to digest. The noise is the acid churning around inside.

Acid, stomach juices and peristalsis (the movement of your intestines as the chyme of food moves along). I'm actually guessing, but I'll be interested to see what others come up with.

It is the acid that we have in our stomach to break the food down. Also our stomachs are like machines and they know roughly what time you eat. If you do not eat at the time it expects it gets grumpy and is basically saying feed me.
