I'm from America. Should I (or can I) book it online before I go to Europe or should I buy it at the station in Prague?

I'm also considering Prague to Budapest instead. Same question applies.

1) Prague -Venice is a loooooooooooooooooong journey.
It takes about 15 hours.. There is one journey a day at 17:14 from the main Prague station and arrives in Venice at 8:45 in the morning..
Alternitively you can break your journey in Vienna (Which is a nice city to visit) There is a night train that goes at 1 in the morning...And catch a train to Venice from there..

As for price.....To Vienna it costs 31 euros one way (22 if you are under 26) And there are discounts for return tickets
I haven't done the full Prague Venice trip but I imagine it would be about 60 euros and no more than 80 (which is price of Prague - Zagreb which is similar length journey)
(That is price of ordinary ticket - You must pay more for a sleeper bed)

2) The journey from Prague to Budapest takes about nine hours and there are two night trains at 11pm and 1am

A one way ticket is 45 euros and about 30 if you are under 26.

This page gives prices (in euro) of more international train journeys from Prague

3)There is no need to book online.. It is easy to book at the station. Any large station has an international window where you book your international tickets..and they speak English..

Most fast inter city routes say that you must reserve a seat..but all that means is buying the ticket before hand rather than on the train (I would give yourself half an hour before your journey for this)
On local (or slow trains) you can ofen pay on the train though the full range of discounts might not be available)

There is no need to worry about places being sold out..but I would go to the station the night before or a few hours before you intend to travel (for night trains) especially if you are traveling in the peak summer period...Also to book a sleeping seat..
Thought personally I prefer an ordinary seat as I find the sleeping cabins claustophobic.. And the seats pull out So unless your cabin is full.. I think the ordinary seat is a more comfy bed!!!
The sleeping couches on the Prague to Budapest especially are likey to get busy.. But on some lines such as Beograd- Tessoulonki you can just go to the ticket inspector on the train and pay an extra 10 euros and get a sleeping cabin that way (You are more likely to get your own cabin than share it..if you buy your bed this way)

NB All international fares have reduction for under 26s and for return ticket..

NB2 There are no border checks within Schenigan zone..but if you cross inbetween ie Austria- Czech..OR From countries out side agreement ie Czech- Hungary.. The border guards will come in your cabin and check your passports at the border.. It is not a big deal controls are pretty perfunctory..but have your passport ready

4)I don't reccomend..booking online. It is not necesary and more expensive...But here is a list of agencies such as raileurope that allow American residents to book online

5)This site here is is a good online timetable for trains all over Europe
