Without an example sentence it's a little hard because "lot of" by itself is not normally used. "A lot of" can be meant literally, i.e. you are referring to an aggregation of objects counted by "lots", but more commonly it just means "many."

"There are a lot of kids running around this house."

You can intensify by placing the word "whole" in front: "There are a whole lot of kids around here."

lot of is more general aspected while a lot of is more/point specific.
