A number of companies leave from Central Station for Brisbane every day. There is a bus booking office at Central which looks after all of them. It is 1000km from Sydney to Brisbane and the bus will take about 14 hours for the trip. It is comfortable enough as long as you are not tall. There is not a lot of legroom in buses.
Most of them travel overnight arriving in Brisbane in the morning. Don't expect to see too much scenery on the way.

There is a train service as well. An overnight train leaves Sydney each evening arriving in Brisbane in the morning. There is a day train that goes as far as Casino in northern NSW and then you connect to a bus that takes you to Brisbane. The trip from Sydney to Casino is in daylight

Yes, very long trip in a bus. Several bus lines will leave from central station in sydney and go to Roma street station in Brisbane. You will be at least 18-19 hours on the bus with only a few 45min breaks to eat. not a lot of fun. Take the plane only $100 more but 1 1/2 hour flight.

There is but I would recommend the train, a bit faster and a lot more comfortable. Or else to travel by plane cost bugger all from Sydney to Brisbane, you might even be luck and get a flight under $100 with Virgin or Jet star
