There are many different regions, each with a different flavor -- it depends what you want to do and see.
Madrid -- the Prado (art) and Retiro Park. and much more.
Bilbao -- the Basque country. See the new Guggenheim. A beautiful city. Basqe culture has some surprises like lots of sheep and bagpipes.
Barcelona -- Beautiful art and architecture. Catalan. A more serious city.
Sevilla -- Andaluz. A much more typical traditional castillian Spanish culture. Moor and Roma (Gypsy) influences.
Santiago de Compostela -- a pilgrim destination for centuries. This is the north western Galician region.
There are also many beach resort cities and islands like Mallorca and Ibiza.

oh god. Lisbone (Lisboa) is the capital city of Portugal!
everything depends on what you look for... the most touristic cities in Spain are Madrid and Barcelona (i like Madrid better than Barcelona). However, i think that all Andalucia (all the south) is warm and its worth it too. the Mediterranean in general is awsome

Sevilla is beautiful.
Granada was amazing, one of my fav. cities. Its got a moroccan culture to it since its close to Africa. Plus the L'Ahambra is there!
Cordoba.... OMG the Mosque over there was breathtaking.
There is Cadiz, the oldest city in Europe. They have an amazing mosque as well down there and the ocean is gorgous.
Barcelona if you are into the arts as well as partying... place is crazy and it doesnt start partying till 12am-1am and lasts well into the morning.
Madrid was a lot of fun wish I had more time there. You can go see a bullfight and actually see them kill it afterwards. Its something to experience, and im not for the killing of the bull, but its not everyday you get to see a bullfight and see the killing of a bull.

Second: Barcelona
Third: Sevilla, Bilbao or Valencia.
Lisbon is not from Spain, it`s Portugal. But also is great!
