I say "in", but a lot of people I know say "on". Maybe it's a regional thing?

In line means the line is moving. On line means it's not, it's a people parking lot, much like waiting for the ticket office or whatever to open.

If your standing on an actual, visible line, than you are standing "on" a line. if you are in a formed line, than you are standing "in" line
