I was about to tell you yes, but now that I think about it the store I did this with was a Borders. Borders (at any rate) has an online option to search nearby stores and pick it up instore. They send you an email when the book is ready. I did this once when I needed a book for a present and I got a Borders about 15 minutes away that had the book ready in less time than it took me to find my keys. So it was really convenient. It's called "search in store", or something. I feel like I may have checked bn.com and they didn't have this feature, but if they do, it's probably called something like in-store pickup, or search nearby stores, or something. You can also try froogle.com on local search putting in your zip and the name of the book - might work (although I find froogle annoying, tends to give you a lot of online sellers when what you want is who has it in neighborhood.)
Really, the best thing to do is call. Much as I hate to admit I shop at chains, I do find Barnes and Noble to be really convenient and their staff really helpful. (my 10% discount from my dad being a 'member' doesn't hurt, either. thanks, dad.) They can check their stock and if they don't have it they can look in their computer and find an area store that does. If nobody has it but they carry the book, they'll just put it on order for you and you can pick it up in the store. (and no, you won't have to pay shipping that way.)

The simplest way is to call the store, or go there, and have them order it for you.
At Border's you can check to see if a nearby store has a book in stock, and f it sisn't ask them to order it -- but I'm not sure you can do it online at Barnes & Noble. Its easier to call the store anyway.

Most bookstores will order a book for you if they don't have it in stock. I don't think you can get it sent to the store and pay for it. You could always look for a "contact us" link at Barnes and Noble and email them and ask.

I'm not sure how, but you can go there and order FROM b&n and they'll call you when they have the book...But while youre at there, I guess you could ask them how it might work online.
