i agree with everyone that said 6-7 weeks by ultrasound. i also read somewhere that they can tell by your levels of hcg hormone.if u have a higher than usual content of the hormones it could mean multiple babies.

Usually when u have blood drawn to check your levels they can guess if u are having multiples by how high the levels are and this can be done very early in the first couple weeks..During ultrasound between 6-8 weeks and up they can detect if more than one baby is in there, they will see 2 sacs,2 babys etc...

Until modern techniques such as ultrasound became a part of antenatal care, most women didn't learn that they were pregnant with twins until they were in labour. But such late surprises are rare today. People typically discover they're having twins during a routine ultrasound, often in the first trimester. Ultrasound is almost foolproof at diagnosing twins, but the greater the number of babies, the less accurate the ability to detect how many there are.

My sister knew she had a high chance of carrying multiples because the original blood test her doctor did on her to see if she was pregnant had a much higher than average level of the hormone the test looks for. She did have twins confirmed by an ultrasound a few weeks later.
