All of the answers have great tips. It does depend on how long your flight is.... if it's 3-5 hours or less you might want to sit by the window to see where you're going if the weather is good. Moving around is less important on shorter flights. But no matter where you're sitting, get up and move around on longer flights.
Drinking water is a good idea for long or short flights... the plane air really dries you out. You can't take liquids from home, but you can buy water in the secure area and take it on board with you.
We wouldn't bother with a blanket even though the airlines don't always provide them anymore.... but that's because we really think you should travel light. Wear a comfortable sweater that you can take off if the cabin air gets too warm or keep on if it's cold.
You can bring your own earphones for movies (if your fight is long enough) if they're compatible with the plane's system or buy them for about $5... sometimes the movies are OK.... sometimes not. Bring books, your own movies and music or whatever to pass the time.

It depends on how long you are flying.
I'm not sure on what the rules are now, but if you can.. bring drinks! There isn't much choice aboard, and you will get thirsty up there!
Also, bring some good headphones for the movie. The ones they provide suck.

Get up and move around the cabin frequently. (that's why it's best to get an aisle seat.)
Do exercises in your seat.
Bring books, music, etc. to pass the time.
I swear by noise-cancelling headphones. They dull the awful, fatiguing sound of the plane.

Some people take sleeping pills with them on long-haul flights, and those inflatable pillows. Blowing your nose case also help with the ear popping.
