a) The main consideration is the question you want to answer.
What is your question? This is the most important part. Why bother to do an experiment if you don't have a question?

b) The next thing is to design an experiment which will answer your question. It will be easiest if it is a yes/no answer. Think careful about possible outcomes and what they will tell you.

c) Next you have to consider which plant species is best suited, though depending on your question this will be obvious.

d) if it is a plant species which has been used in tissue culture before, how was this done? Read the appropriate research papers. Do you have access to the necessary chemicals and plants? Do you have access to the necessary apparatus?

Usually you need a recipe for the agar or nutrient solution. You need to be able to work in a sterile environment. You should have some idea if you have to add plant hormones as they can make a big difference depending what you want to do. You should have ave a place with controlled temperature, light and humidity where you can let the plant tissue grow. While it does this you also have to make sure no bacterial or fungal infection will grow in competition with your plant tissue (unless of course this happens to be part of your experiment). Depending on what you plan to do you may need a dissection microscope with a camera to document your data. You need to have the capability to measure whatever you need to measure in order to answer your question.

You need money to pay for it all or somebody who allows you to use their lab equipment and material. You may need the appropriate training if you haven't used some equipment before.

In addition you should consider providing more detail/ asking a more specific question if you want a more specific answer.
