Do you have your fish already, like you bought them and they are waiting in the bag? In that case, yes, release them immediately (well, float them in the water for 10 minutes first, then open the bags and add a little tank water, and every 10 minutes add a little more tank water until the bag is full - then net the fish and release them into the tank). However, do know that adding aquasafe does not effectively cycle your tank, so you'll nee to do some frequent water changes to ensure the ammonia levels stay in a tolerable range. By frequent, I mean like 10-15% every other day for a couple weeks. Once 2-3 weeks have gone by, you can reduce that to 30% a week.

In the future, please cycle your tank thoroughly.
In the wild, and in established tanks, there are nitrifying bacteria that break down toxic ammonia from fish waste and excess food into less toxic nitrates. In a brand new tank, these bacteria don't exist, so any fish in the tank will produce ammonia, which, not being broken down by bacteria, will kill or weaken the fish. So, it is vital to cycle your tank. For more details, you can google : Fish Tank Nitrogen Cycle

The water is conditioned instantaniously, but the fish need to be aclimated to it.

First, float the bag(s) on the water to equallize the temperature. After half an hour, add tank water to the bags (about 1/2 the volume of the bag, but it does not have to be precise). This gets them used to the water chemistry. After another 1/2 hour, release them.
