The question is: why is she smiling?
I will ask and answer this for you: Leonardo took so much time to paint her that he probably did not take a unique fixed model for his picture, neither a fixed position. The mixture is what makes the Mona Lisa so misterious. She is hapy but angry, sad but relaxed, all at the same time in different days of the process.

She's looking at leonardo & smiling cause leo was telling her how his painting is gonna screw peoples minds in the 21st century.
