Without more details, this is all I can suggest.
--Hydrolic lock - That's more of a 4 stroke symptom. So much fuel in the combustion chamber, the piston can't compress it. Remove the spark plug and kick start it to blow the excess fuel out.
--Check to make sure the top end is/isn't siezed (it's possible the kick start mechanisim is locked up) - remove the left crankcase cover and try to turn the flywheel. If it won't move, then check the top end.
--Remove the exhaust pipe and look in the exhaust port with a flashlight and mirror. Any scoring of the piston, or cylinder wall and the top end will need to be rebuilt.

As already pointed out to you, more info needed.
It could well be that you big end connecting rod bearing on the crankshaft has gone bad, you'll have to pull the top end off to inspect it. That type of seizure is probably more common than the piston to cylinder type.

You're going to have to be a bit more specific. However if the piston is totally dry, that would seen to also indicate no fuel?
