I think it's pretty obvious from reading the manga that Hiko Seijuro is far more powerful than anyone else in the series. The creator even admits he's too powerful to use very often.
After that...I guess

Amakusa Shougo wasn't in the manga, so I don't know about him. But he knows Hiten style, so he's probably towards the top of the list.

Sojiro could have been the strongest but he got mad. Which in turn also made a bit human so he became weaker.
